If someone publically targetted a "JOKE" at anyone I cared about as to their medical condition, any medical condition, I would definitely react. There's a reason in the modern day that many comedians who have historically made a career out of offending humour spend a good chunk of their routine explaining they are just jokes & not to get offended. Ricky Gervais at the Golden Globes is a really good example. "Remember they're just jokes, we're all going to die soon and there's no sequel" Jimmy Carr, Frankie Boyle, Dave Chappelle just some of the recent examples that have done the same in recent routines, there will be far more. You can find the joke by Chris Rock funny or not funny, but ultimately it was just a joke. If we say that joke wasn't appropriate, you pretty much couldn't put together a routine for many comedians. I have little doubt when things settle a bit, some of the cancel culture stuff will target Rock, possibly as much or more as Smith for the slap.
